摘    要:目的建立RP-HPLC测定人血浆中盐酸氨溴索含量的方法。方法选用Hypersil-BDS C18柱(5.0mm×150mm,粒径5μm),流动相:乙腈-pH值7.5磷酸钠缓冲液(65∶35),流速:0.9mL/min,检测波长:248nm,柱温:25℃。结果该方法低、中、高3种浓度的提取回收率为85.2%~97.7%,日内、日间相对标准差为0.77%~12.2%(n=6),在20~640ng/mL范围内线性关系良好。结论本法简便、灵敏、专属性强,能满足含量测定及体内药代动力学研究的需要。

Abstract:Objective To develop a fast reverse-phase high-performance liquid chromatography assay method for the determination of ambroxol hydrochloride.MethodsA Hypersil-BDS C18(5.0 mm×150 mm,5 μm)column was used.The mobile phase consisted of acetonitrile-phosphate buffer(35∶65).Detection wavelength was 248 nm.ResultsA good linear relationship was obtained in the concentration range of 20~640 ng/mL.The average recovery of this method was 85.2%~97.7%.The RSD of within-day and between-day were all less than 15%.ConclusionThe method is simple,rapid and accurate.The result showed that the method was suitable for the determination of ambroxol hydrochloride.
