山楂叶总黄酮预处理对脑缺血再灌注小鼠脑组织抗氧化酶的影响

摘    要:目的:观察山楂叶总黄酮(Hawthorn Leaves Flavonoids,HLF)预处理对局灶性脑缺血再灌注损伤小鼠脑组织抗氧化酶的影响.方法:80只雄性昆明小鼠,随机分为假手术组,缺血再灌注模型组,HLF低、中、高剂量组.采用线栓法建立小鼠局灶性脑缺血再灌注模型,其中HLF低、中、高剂量组小鼠于术前1周分别给与不同剂量的HLF(20、40、60 g/kg).检测脑组织中丙二醛(MDA)的含量、乳酸脱氢酶(LDH)和超氧化物歧化酶(SOD)的活力.结果:HLF低、中、高剂量可明显提高模型小鼠脑组织LDH及SOD活力(P<0.05,P<0.01),HLF中、高剂量可显著降低MDA含量(P<0.05,P<0.01).结论:HLF预处理对局灶性脑缺血再灌注损伤小鼠脑组织有明显的保护作用,其机制与抗氧化有关.

Abstract:Objective: To investigate the effects of Hawthorn Leaves Flavonoids (HLF) pretreatment on antioxidiase activity in brain of local cerebral is chemia reperfusion mice.Methods:80 kunming mice were applied in this study and divided into the following groups at random: sham-operation group, ischemia reperfusion model group and HLF low, middle, high dose group. The local cerebral ischemia reperfusion model was made by suture method.The mice in 3 HLF groups were given different dose of HLF(20,40,60g/kg)l week before operation. The MDA content and LDH, SOD activity in brain were measured respectively .Results: HLF low, middle and high dose could obviously increase the LDH, SOD activity in brain of local cerebral ischemia reperfusion mice (P<0.05, P<0.01); HLF middle and high dose could significantly decrease the MDA content (P<0.05, P<0.01).Conclusions:HLF pretreatment could protect the brain of local cerebral ischemia reperfusion mice, its mechanism may relate to its antioxygen effects.
