摘    要:目的 :探讨阿糖胞苷对体外培养的宫颈癌细胞 (Hela细胞 )端粒酶活性的影响。方法 :采用PCR ELISA法检测经阿糖胞苷处理前后的Hela细胞株端粒酶活性的改变。结果 :经阿糖胞苷处理后 ,Hela细胞的生长受到抑制 ,端粒酶活性明显降低。结论 :阿糖胞苷对Hela细胞具有生长抑制作用 ,其抗肿瘤作用的机理可能与抑制肿瘤细胞的端粒酶活性有关。

Abstract:Objective: To investigate effects to telomerase activity by Ara-c in Hela cells in vitro . Methods: MTT assay was used to investigate the inhibitory effect of Ara-c on Hela cell in vitro . The telomerase activity in cells treated by Ara-c was detected by PCR-ELISA assay which was based upon PCR amplification of the initial telomerase product and detected by ELISA. Results: The growth of Hela cells was inhibited by Ara-c and the telomerase activity in Hela cells was significantly reduced. Conclusion: The Ara-c could directly inhibit the growth of Hela cells. The mechanism of anti-tumor by Ara-c may be explained by the inhibition of telomerase activity in cells by Ara-c.
