UPLC-MS/MS法测定氯霉素滴眼液中氯霉素的含量

目的建立超高效液相色谱-串联质谱法(UPLC-MS/MS)测定氯霉素滴眼液中氯霉素含量。方法采用UPLC BEH C18色谱柱,以乙腈-水为流动相,经梯度洗脱后,利用质谱仪的多反应监测(MRM)模式测定氯霉素含量。结果氯霉素质量浓度在1.0~20.0 ng/ml范围内与质谱相应值呈良好的线性关系,检出限为0.15 ng/ml,定量限为0.5 ng/ml,RSD为0.83%,加标回收率平均值在94.0%~100.3%。结论UPLC-MS/MS法准确度高、灵敏度高、重现性好。

Determination of chloramphenicol in chloramphenicol eye drops by UPLC-MS/MS

Objective To established the method of determination of the content of chloramphenicol in chloramphenicol eye drops by UPLC-MS/MS.Methods The content of chloramphenicol was determined by a multi-reaction monitoring(MRM)mode of the mass spectrometer using UPLC BEH C18 column with acetonitrile-water as the mobile phase and gradient elution.Results The concentration of chloramphenicol in the range of 1.0~20.0 ng/ml showed a good linear relationship with the corresponding values of mass spectrometry.The detection limit was 0.15 ng/ml,the limit of quantification was 0.5ng/ml,and the RSD was 0.83%.The average is range from 94.0%to 100.3%.Conclusion The UPLC-MS/MS method has high accuracy,high sensitivity and good reproducibility.
