摘    要:目的比较盖诺+顺铂加同期照射与单纯照射对非小细胞肺癌细胞凋亡率的影响。方法应用细胞培养技术,分别采用单纯照射,盖诺+顺铂加同期照射应用于人月肺腺癌细胞GLC细胞,进行细胞凋亡实验。通过光镜细胞形态学观察、荧光染色法、流式细胞仪等检测方法观察其诱导凋亡的差异,并进行量化比较效果差异的程度。结果流式细胞仪定量分析细胞凋亡发现,在12h处及时24h处,各处理组凋亡牢之间无显著差异;在48h,同期组中以8Gy的凋亡率最高,盖诺+顺铂加同期照射与单纯照射相比,凋亡率前者明显高于后者,P〈0.05,有显著性差异。结论靠诺+顺铂化疔方案可增加GLC细胞的放射敏感性。盖诺+顺铂加同期照射诱导肺癌细胞发生的凋亡率可明显高于单纯照射应单纯化疗,其值甚至大于单纯照射和单纯化疗所致凋亡指数之和,但这种效应呈现一定的时间-剂量选择性。

Abstract:Objective To compare the effect of lung cancer cell apoptosis induced by radiation or combination of vinorelbine and cisplatin concurrent radiation.Methods Culturing human lung cancer cell GLC to logarithmic growth phase were used for the cell apoptosis test.After treating lung cancer GLC cells with radiation or combination of vinorelbine and cisplatin concurrent radiation and inculating cell for 12 h,24 h and 48 h,the formation of cellular apoptosis was observed by using inverted phase contrast microscope,fluorescent stain and flow cytometery technique.The effects of two treating methods on cellular apoptosis were compared each other.Results In the cell apoptosis test,there was no difference in all groups at 12 h or 24 h except 8Gy at 24 h,while vinorelbine and cisplatin with concurrent radiation could achieve better effect of apoptosis than only radiation at 48 hours(P<0.05).Conclusion Vinorelbine and cisplatin concurrent with radiation could enhance apoptosis sensibility of lung cancer cell induced by radiation.The vinorelbine and cisplatin with concurrent radiation could achieve better effect of apoptosis than radiation or chemotherapy individually.There is selectivity of time-dose on apoptotic peak values of radio-chemotherapy groups.
