iNOS与NO在实验性大鼠慢性细菌性前列腺炎中的表达及意义

摘    要:目的:探讨诱导型一氧化氮合酶(iNOS)与一氧化氮(NO)在实验性大鼠慢性细菌性前列腺炎中的表达及意义。方法:取30只Wistar雄性成年大鼠随机分为对照组和实验组,每组15只。实验组大鼠前列腺内注射大肠杆菌,制备大鼠慢性细菌性前列腺炎模型;对照组大鼠前列腺内注射0.9%无菌生理盐水。1个月后处死两组大鼠取双侧前列腺组织,一侧制片常规HE染色并应用免疫组化法检测iNOS表达,以HPLAS图像分析系统观察其平均灰度。同时采用分光光度计间接比色法检测另一侧前列腺组织NO含量。结果:iNOS在前列腺炎组织的表达比正常组织明显增多(P<0.01),NO在前列腺炎组织中的含量比正常组织明显增多(P<0.01)。结论:iNOS与NO参与前列腺炎的过程,可为前列腺炎的基础与临床研究提供参考。

Abstract:Objective: To investigate the expression of iNOS and NO in chronic bacterial prostatitis rat model.Methods: An experimental CBP model was induced in 15 male Wistar rats by injection of E.coli into the prostate.Other 15 were injected with 0.9% sodium chloride as control group.All the 30 rats were killed after one month and the prostate samples were collected.Immunohistochemistry SP technique was adopted to detect the expression of iNOS and the content of NO was measured by indirect colorimetric method in the prostates.HPLAS high definition image analysis system was used to determine the average of gray-scale of iNOS slices.Results: The expression of iNOS in prostatitis tissues was significantly higher than that in normal prostate tissues(P<0.01),and the content of NO in prostatitis tissues was significantly higher than that in normal prostate tissues(P<0.01).Conclusion: NO and iNOS participates in the prostatitis development,and provides new reference markers for the basic and clinic research of prostatitis.
