电磁脉冲对上皮细胞和小鼠睾丸超微结构的影响

摘    要:目的观察电磁脉冲(electromagnetic pulse,EMP)对中国仓鼠卵巢上皮细胞(Chinese hamsterovary cell,CHO细胞)和小鼠睾丸超微结构的影响,以探讨EMP对生殖系统影响的机制。方法电磁脉冲辐照CHO细胞和BALB/c雄性小鼠,分别于照后6 h、12 h和2 h、12 h取材。制备超薄切片。结果CHO细胞微绒毛减少,溶酶体增多,部分线粒体肿胀,胞浆出现少量空泡;小鼠睾丸组织中细胞间隙加大,有空泡、有髓鞘样结构出现,细胞膜轻度肿胀,溶酶体明显增加,线粒体肿胀、空泡化,内质网和核周间隙均扩张,核内出现空泡、髓鞘样结构。结论电磁脉冲辐照可导致CHO细胞和小鼠睾丸超微结构变化,可能是EMP引起生殖功能改变的原因之一。

Abstract:Objective To observe the effects of electromagnetic pulse(EMP) irradiation on the ultrastructures of CHO cells and mouse testis in order to investigate the mechanisms for procreation.Methods CHO cells and BALB/c mice were exposed to EMP.Samples of the CHO cells and testis were prepared respectively 2h and 12h,and 6h and 12h after the radiation for transmission electron microscopy(TEM).Results In the CHO cell,the number of microvilli decreased,but that of lysosomes increased.Swelling and vacuole were observed in some mitochondria and some necrosis cells.In the murine testicle tissue,distances between cells increased,and there were vacuoles and myelin figures in the cells and nuclei.Some membrane structures were swollen.In some mitochondria,swelling,breaking and vacuole were observed.Lysosomes increased,endoplasmic reticulum(ER) and perinuclear spaces dilated.Conclusion EMP irradiation could induce some changes in the ultrastructures of CHO cells and murine testis,which may infinence mice genital function.
