| 摘 要： | 目的: 建立一套双向凝胶电泳图像分析方法,为下一步差别蛋白的鉴定提供依据.方法:采用一步法分别提取人永生化食管上皮细胞系SHEE和由SHEE转化而来的食管癌细胞系SHEEC的可溶性总蛋白.经双向凝胶电泳分离后,应用Bio-Rad公司的双向电泳凝胶图像分析软件PDQuest 6.2分析SHEE与SHEEC之间可溶性总蛋白的差异表达情况.结果:SHEE和SHEEC细胞的双向电泳图谱经背景消减、点检测和匹配后,获得了各自的差异蛋白点.其中有7个蛋白点只出现在SHEE细胞中,有2个蛋白点只出现在SHEEC细胞中.结论:利用PDQuest 6.2凝胶图像分析软件可快速有效地对两个密切相关的生物系统之间蛋白质样品双向凝胶电泳结果的差别进行分析处理.

ESTABLISHMENT OF A TWO-DIMENSIONAL GEL ELECTROPHORESIS IMAGE ANALYSIS METHOD |

| Abstract: | Purpose: To establish a two dimensional gel electrophoresis image analysis method for analyzing the differentially expressed proteins between SHEE and SHEEC. Methods: The whole soluble proteins of SHEE and SHEEC were extracted by one step and separated using 2 DE. Different expressions of the whole soluble proteins were analyzed using the 2 DE image analysis software, PDQuest 6.2(Bio Rad Inc). Results: After the background subtraction, spot detection and matching, the protein spots which expressed differentially were obtained. Seven protein spots were detected in SHEE, and two protein spots were seen in SHEEC but not in SHEE. Conclusion: The 2 DE results of two intimate biosytems were analyzed quickly and efficiently using PDQuest 6.2 gel image analysis software. |
