三氧化二砷诱导犬髂动脉平滑肌细胞凋亡的实验研究

目的；研究三氧化二砷诱导犬髂动脉血管平滑肌细胞（vascular smooth musclecell，VSMC）凋亡的作用。方法：采用MTT、细胞生长曲线、倒置相差显微镜、细胞爬片HE染色、透射电镜等观察三氧化二砷作用后VSMC的变化。结果：VSMC细胞与三氧化二砷共孵育后出现细胞数目减少，脂肪颗粒增多；电镜等观察到核膜皱缩、核染色质凝结、边集等细胞凋亡现象，并且在一定的范围内此现象随药物浓度的增加而明显。结论：一定浓度的三氧化二砷能抑制VSMC细胞的生长，导致细胞变性，并诱导细胞凋亡。提示三氧化二砷作为涂层支架的药物可以抑制的平滑肌细胞生长，促进平滑肌细胞凋亡。

The Experiment Study on Apoptosis-inducing Effect of Arsenic Trioxide (AsO2) on Canine Iliac Artery Smooth Muscle Cell

Objective:To study the possible apoptosis-inducing effect of arsenic trioxide(AsO2)on canine artery smooth muscle cell. Methods: A wide range concentration of arsenic trioxide were cultured with (vascular smooth muscle cell.VSMC) with different time,cell growth curve, MTT assay, electron microscopy detection, inverted phase contrast microscope, HE staining were used to study cell activity and morphologic changes. Results: After culturing with AsO2, VSMC present cells number decrease,fat granule increase,and apoptotic featuresect. Electron microscopic appearance of VSMC exhibiting membrane integrity, chromatin condensation,apoptotic body formation,ect. Conclusion:AsO2 can induce apoptosis of VSMC
