EphA2与E cadherin在胰腺癌中的表达及其意义

摘    要:目的 探讨EphA2和E-adherin在胰腺癌组织及细胞中的表达及其临床意义。方法 采用免疫组织化学SP方法,检测56例胰腺癌及23例癌旁非肿瘤胰腺组织中EphA2和E-cadherin的表达,并分析其与临床病理因素的关系。采用RT-PCR方法检测EphA2在胰腺癌细胞株BXPC-3,PC-3及PANC-1中的表达。结果 胰腺癌组织中EphA2阳性和E-cadherin阴件的表达率分别为66.07%和57.14%,均显著高于癌旁非肿瘤胰腺组织(P〈0.05);EphA2表达与胰腺癌分化程度、TNM分期和淋巴结转移有关,E-adherin表达与胰腺癌TNM分期和淋巴结有关。在胰腺癌组织中,EphA2与E-cadherin的表达呈负相关;EphA2在胰腺癌向侵袭力细胞株BXPC-3和PANC-1呈高表达,在低侵袭力细胞PC-3呈低表达。结论 EphA2与E-cadherin的表达和/或功能异常可能共同参与胰腺癌的发生发展与转移;联合检测两种蛋白对于胰腺癌转移潜能的判断具有一定的参考价值。

Abstract:Objective To explore the expression and elinieal significance of EphA2 and E-cadherin in pancreatic cancer tissue and cells. Methods The expression of EphA2 and E-cadherin in 56 panereatie carcinomas and 23 adjacent noncancer tissues were studied bv immunohistochemistry, and their relationship to clinicopathological charaeteristies were analyzed. RT-PCR was performed to explore the expression of EphA2, BXPC-3, PC-3 and PANC-1 in pancreatic carcinoma cell lines. Results In pancreatic carcinomas tissuse showed inereased EphA2 expression and reduced E-cad expression which compared with adjacent non-cancer tissues. The expression level of EphA2 had a significant positive relationship with tumor differentiation degree, lymphatic metastasis and elinical stage. However, the expression level of E-eadherin had negative relationship with both the tumor clinieal stage and lymphatic invasion. Furthermore, a significant negative relationship between the expression of EphA2 and E-cadherin was obselwed. The expressions of EphA2 were higher in high-invasive celt lines PXPC-3 and Panc-1 than in low-invasive cell line PC-3. Conclusions The expression and/or abnormal function of EphA2 and E-eadherin may together be involved in the development and progression of pancreatie cancer; the combined measurement of these two proteins may be useful for determination of metastatic potency of pancreatic eareinoma.
