摘    要:目的 探讨人骨形成蛋白(rhBMP-2)对体内,外人脑胶质瘤SHG44细胞周期和增殖的影响。方法 体外培养人脑胶质瘤细胞,绘制rhBMP-2作用前、后的SHG44细胞生长曲线,MTT法测定rhBMP-2对SHG44细胞的增殖影响;以流式细胞仪、电镜和琼脂糖凝胶电泳分别检测其细胞周期、超微结构和DNA片段改变,观察局部注射rhBMP-2对裸鼠皮下SHG44胶质瘤增长的影响,结果 rhBMP-2可抑制

Abstract:AIM To investigate the effects of recombinant human bone mor phogenetic protein (rhBMP 2) on the cellcycle and proliferation of human glioma cells (SHG44) in vivo and vitro . METHODS The human SHG44 glioma cells were cultured in vitro . The growth curve of SHG44 cells with or without rhBMP 2 was plotted. The proliferation effect of rhBMP 2 on SHG44 cells with rhBMP 2 was determined by MTT method. The cell cycle, ultrastructure and DNA fragments of SHG44 cells were detected by flow cytometry, electron microscope and gel electrophoresis agarose respectively. The growth state of SHG44 glioma growing under the skin of nude mice was observed with the local injection of rhBMP 2. RESULTS The rhBMP 2 could inhibit the growth of SHG44 cell in vitro . The DNA content of SHG44 increased in G1 cycle, and decreased in S cycle obviously ( P <0.01). An apoptosis spike (12.3%)was found by FCM assay. The nucleolus chromatin aggregation, bound of aggregation, caryorrhexis and apoptosis cells were observed under the electron microscope. The DNA electrophoresis showed a ladder strap(DNA ladder). SHG44 glioma growth in mice was relatively slower and its volume was smaller with local injection of rhBMP 2 than those without rhBMP 2 injection ( P <0.01). CONCLUSION SHG44 cells could be arrested at G1/S stage and the cell growth could be significantly suppressed by rhBMP 2. The prosess might be carried out through rhBMP 2 induced apoptosis in human glioma cells.
