一氧化氮合酶在豚鼠耳蜗的定位表达

摘    要:目的:研究一氧化氮合酶(NOS)在豚鼠耳蜗的定位分布,以确定NO作为神经递质和血管内皮舒张因子在内耳听觉生理和病理生理机制中的作用。方法:健康纯白红目豚鼠35只,用NADPH黄递酶组化法和NOSmRNA原位杂交技术研究NOS在耳蜗的表达。结果:NOS主要分布于耳蜗螺旋器的内、外毛细胞、螺旋神经节细胞胞浆和血管纹边缘细胞。来源于大鼠小脑的NOSmRNA探针在血管纹无阳性信号,原位杂交能在mRNA水平区别不同来源的NOS。结论:NOS在耳蜗螺旋器有表达,因此NO是听觉传入径路上的神经递质或调质。血管纹边缘细胞能合成释放NO,对耳蜗微循环进行自身调节

Abstract:Objective: To investigate nitric oxide synthase (NOS) expression in cochlea of guinea pig, and define the effect of NO as neurotransmitter and endothelium derived relaxing factor (EDRF) in auditory physiology and pathophysiology of inner ear diseases. Methods: Thirty five white red eyes guinea pigs were used, and expression of NOS in cochlea of guinea pig was studied with NADPH diaphorase histochemistry and in situ hybridization technology. Results: NOS mainly distributed in inner hair cell (IHC), outer hair cell (OHC) of Corti's organ, the cells of spiral ganglion (SG) and the marginal cells of stria vascularis (SV) in the guinea pig cochlea. The NOS in the IHC, OHC and spiral ganglion cell was different from the NOS in the marginal cells of SV. It submitted to brain NOS or endothelium NOS. Conclusion: Expression of NOS in cochlea was detected. It showed NO was neurotransmitter or modulator in cochlea afferent and joined self regulation of microcirculation of the cochlea.
