督脉电针对慢性脑缺血大鼠记忆能力及食欲素受体1表达的研究

摘    要:目的探讨督脉电针对慢性脑缺血大鼠记忆能力及食欲素受体1(OX1R)所产生的影响。方法选用70只Wistar大鼠通过分次结扎双侧颈总动脉法建立慢性脑缺血的大鼠模型,分为4组。造模成功后,将实验大鼠按随机数字表法分为对照组、模型组、立即针刺组、针刺治疗组。对照组不予任何处理,正常饲养;模型组、立即针刺组及针刺治疗组又分为1个月、2个月2小组,每组或亚组的大鼠各10只,立即针刺组首次造模即开始进行督脉电针干预,针刺治疗组在造模开始后第3周开始进行督脉电针干预。采用水迷宫对大鼠进行行为学评价,同时用免疫组织化学法测定OX1R阳性细胞数。结果模型组缺血1个月,大鼠学习记忆能力较对照组减退,2个月时较1个月时有所好转;2个月时针刺治疗组学习记忆能力优于模型组,而立即针刺组略优于针刺治疗组。模型组于造模开始后1个月OX1R表达高于对照组,2个月时进一步增高;1个月时针刺治疗组与模型组相比,OX1R表达差异无统计学意义(P0.05),而立即针刺组与模型组相比,OX1R表达有升高(P0.05);2个月时针刺治疗组与模型组相比OX1R表达有明显升高(P0.05),而立即针刺组与针刺治疗组相比OX1R表达有升高(P0.05)。结论督脉电针干预后促进了脑组织OX1R的表达,同时发现早期干预对OX1R表达的影响优于正常治疗组中OX1R的表达。

The study of Du meridian electroacupuncture on the ability to remember and OX1R expression of rats with chronic cerebral ischemia
Abstract:Objective To explore the effect of Du meridian electroacupuncture on the memory ability and OX1R expression of mice with chronic cerebral ischemia.Methods The cerebral ischemic models were established by fractionally ligating bilateral carotid arteries.Seventy mice were divided into four groups by random number table method:control,model,immediately acupuncture and acupuncture groups.The behavior of the models was evaluated by water maze.The number of OX1R positive cells was determined by immunohistochemical.Results After 1 month ligation,the memory ability of mice was impaired in model group 1 month later,and improved after 2 months .The memory ability in acupuncture group was better than that in model group,immediately acupuncture group was better than that in acupuncture group,and there was significant difference.The expression level of OX1R of model group was higher than that in control group,and the expression of OX1R was further increased after 2months.There were no significant differences between the model group and the acupuncture group at 1 month,but the expression of OX1R in the immediate acupuncture group was higher than that of model group.The OX1R expression in acupuncture group was higher than model group,and the same result between immediate acupuncture group and acupuncture group after 2 month.Conclusions The Du meridian electroacupuncture promote the brain tissue OX1R expression of mice with chronic cerebral ischemia.
