摘    要:目的:探索益智中不同类别指标性成分的含量测定方法,并测定不同种植基地圆柚酮、杨芽黄素和益智酮甲的含量。方法:采用固相萃取和高效液相色谱法测定益智果实中圆柚酮、杨芽黄素和益智酮甲的含量。色谱柱为菲罗门C6苯基柱(250×4.6mm,5μm),以乙腈和水为流动相,流速1.0mL/min,柱温30℃,检测波长240nm。结果:该方法线性,精密度,稳定性均良好,圆柚酮、杨芽黄素和益智酮甲的平均回收率为98.61%-99.98%。不同种植基地中圆柚酮的含量比较接近,但是杨芽黄素和益智酮甲的含量差异较大。广西和广东产的益智中杨芽黄素含量较低,益智酮甲含量较高。海南产益智中杨芽黄素含量较高,益智酮甲含量较低。结论:该方法准确可靠,可用于益智药材及相关产品中圆柚酮、杨芽黄素和益智酮甲多指标成分的含量测定,为完善益智药材的质量标准提供参考。

Determination of three compounds in Alpinia oxyphylla by SPE-HPLC

Abstract:Objective:To explore a characteristic chemical marker of Alpinia oxyphylla(A.oxyphylla)and to establish a method to determine the contents of nootkatone,tectochrysis and yakuchinone A in fruit of A.oxyphyllacollected from different sources.Method:The contents of nootkatone,tectochrysis and yakuchinone A were determined by SPE-HPLC.HPLC analysis was made on a C6-phenyl column(4.6mm ×250mm,5μm)with acetonitrile and water.The detection wavelength was 240 nm.Results:The HPLC method showed good linearity,precision and stability.The average recovery rates of three compounds were 98.61%to 99.98%.The content of nootkatone was similar at different sources.The contents of tectochrysis and yakuchinone A showed obvious difference among the plants collected from different sources.The content of yakuchinone A was high at Guangdong and Guangxi harvest zone and the content of tectochrysis was high at Hainan harvest zone.Conclusion:The method is accurate and reliable.It is appropriate for the quantitative determination of nootkatone,tectochrysin and yakuchinone A in A.oxyphylla and its production.
