PWM激活的淋巴细胞亚群的功能研究

摘    要:用放射性标记化合物掺入法研讨PWM诱导人血淋巴细胞亚群的功能及其调节。实验表明,PWM细胞对LPS活的B细胞转化有促进作用,受40Gy辐照后,它的作用显著下降,但仍有一定加强效应。PWM细胞对PWM激活的淋巴细胞转化的促进作用不明显,对ConA和PHA激活的细胞无效。用玫瑰花环法分离T、B细胞,用CD4、CD3和B3株单克降抗体(McAb),以亲和层析和Panning法分离各淋巴细胞亚群,分别研究PWM对各亚群的激活及相互调节作用.结果表明:B细胞被PWM激活强于T细胞,PWM对B和非CD4细胞激活作用相当,CD2细胞次之,CD4细胞最弱.T细胞或CD4细胞与B细胞均有协同作用,前者作用更大.PWM-非CD4和PWM—B细胞无协同作用,CD6细胞与B细胞有抑制作用。

Abstract:he function and regulation in PWM-induced human blood lymphocyte subsets were studied by incorporation method of radioisotope labelling This study found the enhanced effect of PWM- induced cells on human blood B lymphocytes stimulated by LPS After exposed to 40Gy the enhancedeffect decreased significantly ,but it still had certain effect on B lymphocyets,for the effect Of PWM-induced cells on lymphosytes stimulated by PWM there is no evidence,and the effect of PWM- induced cells on lymphocytes on lymphocytes stimuated by Con A and PHA did not exist.T and B lymphocytes were separated by rosetting procedures and three lmphocytes were separated subsets were separated by CD4, and B monoclonal antibodies ,The effect of stimulation and regulation oflymphocyte subsets wal studied respectively ,This paper reflected that PWM can stimulate both T and B cells but B cells were more sensitive to the same as thatOf non-CD4 cells. The response of CD3 Cells tO PWM was lower,and the CD4 cells was the lowest.Both T and CD4 lymphocytes were synergistic to B lymphocytes but such a kind of synergistic effect in T cells more obvious than that in CD4 group The suppressive effect of CD8 cells on B cells was found .
