细胞周期因子cyclinD1,cyclinE,P27KIP1在肝细胞癌中的表达研究

目的探讨cyclinD1,cyclinE,P27KIP1在肝细胞癌中的表达及其意义.方法应用免疫组织化学S-P法检测50例肝细胞癌组织标本及44例相应的癌旁肝组织标本中的cyclinD1,cyclinE,P27KIP1的表达.结果cyclinD1,cyclinE,P27KIP1在肝细胞癌组织中的阳性表达率均高于癌旁组织(P<0.05).P27KIP1的低表达,cyclinD1的过度表达与肝细胞癌临床分期、有无转移显著相关.cyclinE过度表达者也易发生远处转移.肝癌组织中P27KIP1的低表达与cyclinE的过度表达有相关关系.P27KIP1阳性表达者术后生存率高.结论P27KIP1、cyclinD1作为调节细胞周期的重要因子,在肝细胞癌的发生、发展中发挥重要作用.P27KIP1是预测肝癌转移和预后的一个指标.

Expression of cyclinD1,cyclinE and P27KIP1 in hepatocellular carcinoma

Objective:To investigate the expressions of cyclinD1,cyclinE and P27 KIP1 in hepatocellular carcinoma and their relationships with clinicalpathology.Methods:Expressions of cyclinD1,cyclinE and P27 KIP1 were tested by S-P immunohistochemistry in 50 cases of hepatocellular carcinoma(HCC) and 44 cases of adjacent noncancerous tissues.Results:It was found that positive expressions of cyclinD1,cyclinE,P27 KIP1 in hepatocellular carcinoma were higher than in adjacent noncancerous tissues.The expressions of cyclinD1 and P27 KIP1 had a significant correlation with intrahepatic metastasis and clinical stage.CyclinE overexpression also had positive relationship with intrahepatic metastasis.There was a significant relationship between P27 KIP1 and cyclinE expression.Kaplan-Meier survival analysis showed that patients with P27 KIP1 expression had a high overall survival rate.Conclusion:Being important cell-cycle modulators at the G1-S boundary,cyclinD1 and P27 KIP1 may contribute to carcinogenesis and progression of HCC,P27 KIP1 expression has prognostic significance.
