甲基强的松龙和脑源性神经生长因子联合治疗促进大鼠脊髓损伤后髓鞘再生和功能恢复

摘    要:目的:研究甲基强的松龙(MP)和脑源性神经生长因子(BDNF)联合治疗对大鼠脊髓损伤后髓鞘再生和功能恢复的影响。方法:采用28只SD大鼠T10脊髓损伤模型,随机分成MP和BDNF治疗组(A组),MP和脑脊液(CSF)对照组(B组),每组14只。应用逆转录-聚合酶链反应(RT-PCR)及免疫组织化学方法观察MP和BDNF联合治疗对脊髓损伤后髓鞘再生的作用,同时采用脊髓运动功能(BBB)评分观察脊髓神经功能的恢复情况。结果:伤后1d,A组中髓鞘蛋白前脂蛋白(PLP)mRNA的表达较B组增加了45.3%,差异有显著性意义(P<0.05)。伤后10周,A组中快蓝(LFB)和髓鞘碱性蛋白(MBP)阳性的神经髓鞘的表达较B组增加了2倍,差异有显著性意义(P<0.05)。A组大鼠脊髓神经功能恢复较好。结论:MP和BDNF联合治疗对大鼠脊髓损伤后髓鞘再生和功能恢复有明显的促进作用,是急性脊髓损伤一种有效的联合治疗方案。

Abstract:Objective To investigate the effects of combination therapy with methylprednisolone (MP) and brain derived neurotrophic factor (BDNF) on remylination and neurological recovery of function after spinal cord injury in rats. Methods Contusion injury to rat spinal cord was produced at T 10 vertebra level. Twenty eight rats were randomly divided into two groups: Group A received MP and BDNF; Group B received MP and CSF. The spinal cord neurofunctional recovery was observed by using RT PCR, immunohistochemical method and BBB. Results One day after therapy, PLP mRNA expression in the Group A resulted in a 45.3% increase compared with that in the Group B ( P <0.05). The expressions of LFB and MBP positive myelin in the Group A were increased twice more than that in the Group B 10 weeks later ( P <0.05). The neurofunctional recovery was in a good condition in the Group A. Conclusions Combination therapy with MP and BDNF promotes significantly remyelination and functional recovery following spinal cord injury and is an effective approach to treatment of spinal cord injury.
