牛磺酸抗肝脏缺血再灌注损伤的实验研究

摘    要:为观察牛磺酸对肝脏缺血再灌注损伤的保护作用,建立SD大鼠肝血再灌注模型,将大鼠分成5组,分别于缺血前经外周静脉、门静脉注入牛碘酸及再灌注前经门静脉注入牛磺酸,及假手术组组。结果表明,缺血前给牛磺酸组具有抑制缺血再灌注时肝细胞内酶的漏出,降低肝组织内丙二醛(MDA)含量,提高超氧化歧化酶(SOD)、过氧化氢酶(Cat)、谷胱甘肽过化物酶(GSH-PX)、Na、KATP酶及Ca^2+-ATP酶活力,抑

Abstract:To study the protective effect of taurine on hepatic ischemia/reperfusion injury, the models of the liverischemia and reperfusion in Spraque-Dawley rats were made. The rats were devided into 5 groups. Taurine was intra-venously administered before hepatic ischemia and injected into portal vein be fore hepatic ischemia and reperfusion, re-spectively, These rats were cornpared with control and false groups to evaluate the levels of ALT and LDH in serum be-fore and after ischemia, and the levels of superoxide dismutase (SOD),catalase (Cat), glutathione peroxidase (GSH-PX),Na+,K+-ATPase, Ca2+-ATPase, malonyldialdehyde (MDA) in liver tissues and cellular and mitochondrial calcium be-fore and after reperfusion. The results showed that the elevations in serum ALT and LDH as well as MDA 1evels in livertissues were markedly suppressed before ischemia or reperfusion, the levels of SOD, Cat, GSH-PX, Na+,K+-ATPaseand Ca2+-ATPase were significantly increased, and cellular and mitochondrial calcium levels were markedly reduced inthe groups administered with taurine, respectively. Those levels showed significant difference (P<0. 05) when com-pared with control group. This study shows that taurine has marked effects of protecting the liver against ischemia-reperfusion injury and improving liver function.
