反相高效液相色谱法测定脑脊液中头孢三嗪浓度

目的建立脑脊液中头孢三嗪含量的测定方法。方法以C18色谱柱、0.05mol.L-1Na2HPO4(pH=7.20)∶甲醇=85∶15为流动相,在35℃2、40nm波长下分离、检测头孢三嗪,外标法定量。结果线性范围为0.625~20.0mg.L-1,r=0.9989。10.0、5.0、1.0mg.L-1头孢三嗪的平均回收率为98.5%。3浓度的平均日内精密度(RSD)为3.60%,平均日间RSD为4.06%。结论本方法快速、简便、准确,可用于科研和临床工作中头孢三嗪脑脊液浓度的快速检测。

Determination of ceftriaxone in human cerebrospinal fluid by RP - HPLC

Objective A reverse phase high perfermance liquid chromatography(RP-HPLC) method is established for the determination of ceftriaxone in human cerebrospinal fluid.Methods Ceftriaxone wes separated on a C_(18) column at 35℃ using a mobile phase of methanol-0.05 mol·L~(-1)Na_2HPO_4 (pH=7.20)(15∶85), and was detected at 240nm. Results The calibration cure was linear in the range from 0.625 to 20.0 mg·L~(-1) with r=0.9989. The RSD within a day were (2.29%)～5.24%(the average value was 3.60%),and the RSD between days were 2.20%～6.41%.The average recovery of ceftriaxone was 98.5%.Conclusion This method is simple and accurate,and could be used for the determination of ceftriaxone in human cerebrospinal fluid in clinic and scientific researches.
