抗细胞间粘附分子-1单克隆抗体对哮喘肺组织T细胞增殖能力的影响

摘    要:目的:探讨抗细胞间粘附分子-1(ICAM-1)单克隆抗体(单抗)对过敏性小鼠哮喘模型肺组织T淋巴细胞增殖反应和活化状态的影响,方法:应用鸡卵清蛋白致敏和刺激小鼠以复制过敏性哮喘模型,收集肺组织T细胞并以流式细胞仪检测其增殖反应,以及活化后产生白细胞介素-5(IL-5)的能力。结果:哮喘小鼠肺组织T细胞的增殖反应明显增强,肺组织局部所产生的IL-5的水平也显著增高,经抗ICAM-1单抗处理后,T细胞增殖能力显著降低,同时,抗ICAM-1单抗还可以抑制IL-5的产生。结论:抗ICAM-1单抗能够抑制肺组织局部T细胞的增殖以及IL-5的产生。

Abstract:Objective:The aim of the present study was to explore the effects of anti-intracellular adhesion molecule-1(ICAM-1) monoclonal antibody (mAb) on proliferation of T lymphocytes and IL-5 production in sensitized mice lungs, and to futher elucidate the role of ICAM-1 in the pathogenesis of bronchial asthma. Methods: Female BALB/c mice were sensitized and challenged with ovalbumin to prepare animal asthma model. Flow cytometry was used to determine proliferation response of lung T cells, and interleukin-5 concentration in homogenized supernatant of lung tissue were determined by ELISA. Effect of anti-ICAM-1 mAb on proliferation of T lymphocytes and IL-5 production were observed. Results: In sensitized mice challengend with ovalbumin 20 minutes once a day for 6 days, the number of proliferating T cells in lung tissue was much higher that that in control mice. In mice treated with intravenous injection of anti-ICAM-1 mAb before each challenge, proliferating T cells dereased by 54%.the results also showed that anti-ICAM-1 mAb prevented antigen-induced production of interleukin-5 in lung tissue.Conclusions:Anti-ICAM-1 mAb is able to inhibit antigen-induced proliferation of pulmanary T cells as well as productions of interleukin-5. These data suggested that the blockade of antigen-presenting cells'functions in airways could be of value in treatment of human asthma.
