摘    要:目的采用高效液相色谱法(high performance liquid chromatography,HPLC)测定山核桃外蒲壳中4,8-二羟基-1-四氢萘酮的含量。方法采用HPLC,色谱柱为Shim-pack C18(250mm×4.6mm,5μm),以甲醇-水(40∶60)为流动相,流速为0.8ml/min,检测波长为333nm,柱温为25℃。结果 4,8-二羟基-1-四氢萘酮在1.13~20.34μg/ml范围内与峰面积呈良好线性关系,r=0.999 9,平均回收率为101.8%,相对标准偏差为1.21%(n=6)。结论所建立的HPLC准确可靠、结果稳定、重现性好,可用于山核桃外蒲壳4,8-二羟基-1-四氢萘酮的含量测定。

Abstract:Objective To establish a high performance liquid chromatography (HPLC) method for determi ning the contents of 4, 8-Dihydroxy-l-tetralone in the hull of Carya cathayensis Sarg. Methods The HPLC determination was carried out on a Shim-pack C18 (250 mm X 416 ram, 5 /μm) column with mobile phase of methanol-water (40:60), flow rate of 0.8 ml/min, column temperature of 25 ~C, and detection wave length of 333 nm. Results The concentration of 4,8-dihydroxy-l-tetralone had a good linear relation- with the peak area within the range from 1.13 to 20.34 /μg/ml (r=0. 999 9). The average recovery was 101.8%, and relative standard deviation was 1. 21G (n = 6). Conclusion The established HPLC method can be used for determining the contents of 4,8-dihydroxy-l-tetralone in the hull of Carya cathay- ensis Sarg because of its accuracy, reliability, stability, and good reproducibility.
