小鼠TIL,LAK细胞穿孔蛋白mRNA表达与其抗瘤作用的关系

摘    要:用原位杂交法检测小鼠不同培养时期的TIL和LAK细胞中穿孔蛋白(PFP)mRNA的表达,并用显微图象分析系统处理检测PFPMRNA表达水平。结果表明:TIL细胞的PFPMRNA表达在第14、21天时达高峰,28天时开始下降;而LAK细胞则在第67天时即达到较高水平,21天时开始下降,但TIL细胞的表达水平显著高于LAK。将同时期的TIL及TAK细胞进行体内外抗瘤活性测定,发现两者的PFPmRNA水

Abstract:The level of PFP mRNA expression in TILs and LAK cells of mouse during various culture time was investigated with hybridization in situ and computer-microphotography analysis system. The results showed that the quantity of PFP mRNA expression in TILs increased to the highest level on cultured 14th to 21th day, and then declined to lower level on 28th day, nevertheless, that in LAK cells got to higher level on 7th day and then dropped to lower level on 21st day, In addition, the level of PFP mRNA expression in TILs was remarkably higher than that in LAK cells, Meanwhile, we detected the antitumor activity of these TILs and LAK cells in vivo and in vitro, It was found that there was significant correlation between the level of PFPmRNA expression and the cytotoxicity in vitro in TILs as well as LAK cells, but there was no notable correlation between the cytotoxicity in vitro and antitumor activity in vivo of both TILs and LAK cells, According to above results, it may be inferred that PFP plays an important role in cytotoxicity of TILs and LAKcells, and the antitumor activity in vivo of TILs and LAK cells is affected by various factors besides PFP.
