黄芪建中汤抑制脾气虚证肺癌小鼠肿瘤转移的实验研究

目的:探讨黄芪建中汤对脾气虚证肺癌小鼠抑制肿瘤转移的作用机制。方法:苦寒泻下法建立小鼠脾气虚证Lewis肺癌模型,模型成功建立后,按实验设计方案处理各组小鼠,同时观察小鼠的整体情况。14 d后,眼球取血,检测血清木糖水平,28 d后,取胸腺、肺及肿瘤组织。分别称取每只小鼠胸腺重量、体重,计数肺转移结节数,计算胸腺指数,采用RT-PCR方法检测nm23、CD44的表达。结果:脾气虚小鼠的整体状况及免疫功能降低,高剂量黄芪建中汤可明显改善脾气虚证Lewis小鼠的整体情况,升高小鼠胸腺指数,结节数少于模型组;黄芪建中汤各剂量组均可下调CD44-mRNA的表达、上调nm23-mRNA的表达。结论:高剂量黄芪建中汤对脾气虚证肺癌小鼠肿瘤的转移具有一定的抑制作用能够,其机制可能与提高胸腺指数,下调CD44-mRNA及上调nm23的表达有关。

Experimental Study on Inhibition of Huangqi Jianzhong Decoction on Metastasis in Mice With Lung Cancer of Deficient Syndrome of Spleen Qi

Objective：To study inhibition and mechanism of Huangqi Jianzhong decoction on metastasis in mice with lung cancer of deficient syndrome of spleen Qi.Method：Lewis lung cancer of deficiency syndrome of spleen Qi models were established by cold and bitter rhubarb lax method in mice.14 d later,general states of mice were observed,content of xylose in serum was determined.28 days later,thymus,lung and tumor tissue of mice were taken out.Body and thymus weight were detected.Expression of nm23,CD44 were detected by RT-PCR.The number of pulmonary metastatic nodules and index of thymus were calculated.Results：The general states and immune function of mice in model group were lower than those in normal group.Compared with model group,general states were improved,index of thymus was increased,number of pulmonary metastatic nodules was decreased in Huangqi Jianzhong decoction high dose group.Expressions of CD44-mRNA were lower and expressions of nm23-mRNA were higher in Huangqi Jianzhong decoction groups than those in model group.Conclusion：High dose Huangqi Jianzhong decoction has inhibition effect on metastasis in mice with lung cancer of deficient syndrome of spleen Qi.Its mechanism may be related to increasing index of thymus,up-regulating nm23 expression,down-regulating CD44-mRNA expression.
