摘    要:目的探讨曲美他嗪对大鼠心力衰竭(心衰)模型心肌能量代谢的干预作用。方法雄性SD大鼠随机分为4组,腹主动脉缩窄法制备SD大鼠心衰模型,造模后4周分别给予双蒸水或不同剂量曲美他嗪灌胃,连续4周。心脏超声测定左室舒张期内径(LVD)和收缩期内径(LVS)、左室射血分数(LVEF)和短轴缩短率(FS);电镜观察心肌线粒体结构变化;放射免疫法检测游离脂肪酸(FFA)。结果与模型组比较,曲美他嗪高、低剂量组LVS、LVD均明显缩小,LVEF、FS均升高,FFA水平均降低。结论曲美他嗪能改善心衰模型大鼠心肌能量代谢,改善心功能。

Abstract:Objective It is to explore the intervention of trimetazidine on the energy metabolism of cardiac muscle in rat models with heart failure.Methods Male SD rats were randomly divided into 4 groups,and rat models with heart failure were established by coarctation of abdominal aorta.In 4 weeks after the models were established,every group was treated with double distilled water or different dosage of trimetazidine by intragastric administration for 4 weeks.LVD,LVS,LVEF and FS were detected by heart ultrasound,the changes of construction of myocardial mitochondria were observed by electric microscope,FFA was determined by radio-immunoassay.Results Compared with model group,LVS,LVD were both obviously shorter,LVEF and FS were higher,the level of FFA was lower in high and low dosage of trimetazidine groups.Conclusion Trimetazidine can improve energy metabolism of cardiac muscle and heart function in rat models with heart failure.
