注射D-半乳糖致大鼠衰老效应的评价

摘    要:目的评价注射D-半乳糖(D—galactose)的致衰老效应。方法30只雄性Wistar大鼠随机分为对照组、模型1组和模型2组。每组各10只。模型1组每天皮下注射D一-乳糖50mg/kg:模型2组每天腹腔注射D-半乳糖500mg/kg;对照组每天颈部皮下注射等量(50mg/kg)生理盐水。各组大鼠均连续给药6周。观察各组体重增加比例、胸腺指数、脾指数、超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量。结果与对照组比较,模型1组和模型2组体重增加比例降低,胸腺明显萎缩,胸腺指数降低(P〈0.01);胸腺组织出现明显病理变化;血清中SOD活性明显下降(P〈0.01)。MDA含量明显升高(P〈0.01)。结论皮下注射和腹腔注射D-半乳糖诱导的衰老效应基本相似。

Abstract:Objective To evaluate the aging effect induced by injection of D-galactose (D-galactose). Methods 30 male Wistar rats were randomly divided into control group, model 1 group and model 2 group, every group contained 10 rats. Rats in model 1 group were injected 50 mg/kg D-galactose subcutaneously once daily, and rats in model 2 group were injected 500 mg/kg D-half lactose intraperitoneally once daily. Rats in control group were injected saline subcutaneously once daily. Rats in each group had continuous administration for 6 weeks. Then the gain weight ratio, thymus index, spleen index, serum superoxidc dismutase (SOD) activity, malondialdehyde (MDA) content in rats of the three groups were observed. Results Compared with the control group, rats in model 1 group and model 2 group demonstrated decreased gain weight ratio, sbrinked thymus and lower thymus index (P 〈 0.01); pathological changes in thymus were observed; SOD activity significant reduced (P 〈 0.01), and MDA content significantly increased (P 〈 0.01). Conclusion The aging effect induced by subcutaneous and intraperitoneal injection of D-galactose is similar.
