摘    要:目的: 观察四氧嘧啶型糖尿病大鼠颌下腺组织学变化及糖尿病对神经生长因子(NGF)在颌下腺表达的影响.方法: 雄性Wistar 大鼠20只,对照组5只,实验组15只.用四氧嘧啶复制糖尿病大鼠动物模型, 1、2、3个月时,取大鼠颌下腺HE染色光镜下观察,用免疫组织化学方法观察NGF在颌下腺的表达.结果: 随着糖尿病加重,颌下腺出现组织萎缩,腺泡缩小,形态不规则等变化.不同月龄糖尿病大鼠NGF表达阳性与对照组比较,呈下降趋势.结论: 糖尿病状态下,颌下腺出现部分腺泡萎缩等变化,且随病程的延长而显著;NGF的表达逐渐降低,说明颌下腺颗粒曲管细胞合成和分泌NGF的功能降低,而NGF缺乏与糖尿病神经病变的发生发展有密切关系.推测实验性糖尿病对大鼠颌下腺结构和功能有一定损害作用.

Abstract:Objective: To investigate the histological changes and expression of nerve growth factor(NGF) in submandibular gland of alloxan diabetes rats. Methods: Masculinity Wistar rats were divided into control group(n=5) and experiment group(n=15).The diabetes rats model was induced by injecting alloxan into peritoneal cavity.1,2 or 3 month after that,we observed the rat submandibular gland under light microscope after HE pigmentation, and used the immunohistochemistry method to observe the NGF expression in the submandibular gland.Results: With the development of diabetes,there appeared tissue atrophy,diminutival acini and their irregular shapes,and other changes in the submandibular gland.The NGF masculine cells in the submandibular gland were less than the age-matched control group,showing a descend tendency with the progression of diabetes.Conclusion: In diabetes,the acinus atrophy and other changes in the submandibular gland become remarkable as the prolonging of the disease course;the gradually decreased expression of NGF demonstrates NGF synthesis and secretion was decreased in the granular convoluted tubule of the submandibular gland.The lower NGF levels may be associated with the development of diabetic neuropathy.Therefore,it is inferred that the experimental diabetes may do some damage to the structure and the function of the rat submandibular gland.
