摘    要:摘要] 目的 应用计算机辅助精液分析仪(CASA)检测rhTNF-α孵育前后及应用黄芪进行体外干预前后精子运动参数的变化,以期探讨药物干预的可能性。方法 34例健康成年男性自愿者手淫法获得精液,符合WHO精液正常标准者26例进入实验流程。按Percoll梯度离心法制备成为精液标本,将精子密度调整为10×106/ml,再从26份经过处理后的精液标本中各取4.5 ml分装于3支无菌试管(1.5 ml/管),即:空白对照组,实验对照组,干预组,78份标本。空白组不加rhTNF-α,实验组和干预组试管分别加入终浓度270 pg/ml的rhTNF-α以后,再向干预组试管加入终浓度为10 mg/ml的黄芪注射液,然后将所有试管置于37℃、5% CO2培养箱孵育2 h后分别取出10μl应用CASA进行精子活力分析。结果 应用黄芪进行体外干预后,空白对照组,干预组中运动参数均明显高于实验对照组,差异有统计学意义(p﹤0.01或p﹤0.05)。结论 黄芪可明显提高精子的活力,应用黄芪进行体外处理精液获取优质精子具有潜在的临床价值。 关键词] 精子 重组人肿瘤坏死因子-α 凋亡率 黄芪

Research of inhibitory action on sperm of rhTNF-a interfered by Radix Astragali
Abstract:Abstract] objective: To probe into the possibility of intervention by medicine, the movement parameters of sperm and sperm incubated with rhTNF-a and the sperm added radix astragali after incubated with rhTNF-a in vitro were detected by CASA. Method:Thirty-four semen samples collected by masturbation were analyzed according to WHO protocol. Semen samples from twenty-six healthy men are prepared by percoll gradient centrifugation. Sperm suspension was diluted to a concentration in 10×106/ml by Ham’s F10 medium. Take 4.5ml samples from each of the semen samples handled, put them into three germ free test tubes(1.5ml/T),78 tubes in all, that is ,three groups: control group, experiment group and intervention group, 78 samples. No rhTNF-a added into the control group, and rhTNF-a in 270pg/ml final concentration added into the experiment and intervention groups and 10mg/ml Radix Astragali added into the intervention group according to the rate at 1:1, put all of the samples into the incubator to incubate for 2 hours at 37℃, in 5% CO2. Taking of samples: take out of 10µl incubated semen from each of the samples to analyze the spermatozoa by CASA. Result: All the samples were detected by CASA after Radix Astragali added into, the significant differences were found between experiment group and control group and intervention group in viability, sperm activity(A+B), straight line velocity, curve liner velocity, average path velocity of the sperm. The movement parameters of sperm in the control group and intervention group were higher than that in the experience group(p<0.01 or p<0.05). Conclusion: Radix Astragali can obviously improve the sperm activity, using Radix Astragali to treat with sperm to get High-quality sperm is of potential Clinical Value.
