毛花甙丙抑制氧诱导乳鼠视网膜新生血管的实验研究

摘    要:目的 探讨毛花甙丙对乳鼠氧诱导视网膜病变模型中视网膜新生血管的抑制作用。方法 将20只生后第7天的C57乳鼠经体积分数75% ±1%的氧诱导5d后,分别给予左眼玻璃体内注射0.50μg(10眼)和0.10μg(10眼)毛花甙丙注射液,右眼注射相同剂量的磷酸盐缓冲液做对照,在生后第17天时进行视网膜铺片染色,计算视网膜新生血管面积和视网膜无灌注区面积,双眼间做配对样本t检验。结果 给予0.05μg和0.10μg毛花甙丙玻璃体内注射后视网膜新生血管的面积分别为(1.09±0.32)mm2和(0.92±0.40)mm2,明显低于对照眼的(1.84±0.59)mm2和(2.03±0.35)mm2(均为P<0.05);0.05μg、0.10μg毛花甙丙注射后视网膜无灌注区面积分别为(1.40±0.44)mm2 和(1.07±0.40)mm2,明显低于对照眼的(2.00±0.65)mm2和(1.83±0.46)mm2(均为P<0.05)。同时0.10μg毛花甙丙治疗效果优于0.05μg。结论 毛花甙丙对氧诱导视网膜病变乳鼠模型中视网膜新生血管的生长具有抑制作用,同时还能促进视网膜无灌注区的修复。

Abstract:Objective To investigate the inhibitive effects of digilanid C on retinal neovascularization in the oxygen induced retinopathy of mice. Methods Sevenday-old C57 BL/6 mice were exposed t0 75% + 1% oxygen for 5 days , and then returned to room air to induce retinal neovascularization. Twenty mice were divided int0 2 groups equally. 0. 05 yg and 0. 10 yg digilanid C were intravitreally injected into the left eye of each mouse in respective groups on postnatal day 12. Phosphate buffer saline was intravitreally injected into the right eye as control. Retinal neovascularization was evaluated by isolectin GS stairung of retinal blood vessels on P17. The area of neovascularization and obliteration were calculated, and the difference between digilanid C eyes and the PBS eyes was analyzed by the paired t test. Results In both the 0. 05 yg group and 0. 10 pdg group, the area of neovascularization in digilanid C eyes were ( 1. 09 + 0. 32 ) mmz and (0. 92 +0. 40) mm2 ,respectively , were significantly lower than that of the PBS eyes , which were ( 1. 84 + 0. 59 ) mm2 and ( 2. 03 + 0. 35 ) mm2 , respectively , there were statistical differences ( all P < 0. 05 ) . In both the 0. 05 pdg group and 0. 10 Vg group , the area of obliteration in digilanid C eyes were ( 1. 40 + 0. 44 ) mm2 and ( 1. 07 + 0. 40 ) mm2 . respectively , were significantly lower than that of the PBS eyes , which were ( 2. 00 + 0. 65 ) mm2 and ( 1. 83 + 0. 46) mm2 , respectively , there were statistical differences ( all P < 0. 05 ) . At the same time ,the inhibitive effect of 0. 10 Vg group was better than that of 0. 05 pdg group. Conclusion Digilanid C inhibits the retinal neovascularization in a mouse model of oxygen-induced retinopathy , and also can promote the repair of obliterative area.
