不同浓度KH902对兔碱烧伤后角膜新生血管的抑制作用

摘    要:目的 观察不同浓度KH902对兔碱烧伤后角膜新生血管(cornealneovascularization,CNV)的抑制作用。方法 取成年新西兰兔24只,建立兔碱烧伤模型,将兔随机分为4组,使用KH902眼用注射剂(康柏西普)配置不同浓度的KH902,A组2.5mg·mL-1,B组5mg·mL-1,C组10mg·mL-1,D组为对照组(生理盐水),均为每日滴眼3次。分别在制模后3d、7d、14d、30d拍摄眼前节照片,观察CNV情况并行角膜激光共焦显微镜检查,并计算CNV面积。观察各组处死兔常规病理和VEGF表达情况。结果 在制模后28d内,四组CNV形成,其中D组CNV较A、B、C组生长明显;对各组切片进行观察发现,四组均出现了角膜上皮及基质细胞的损失和修复、炎性细胞的浸润和CNV的生长,但D组的炎性细胞较密集,CNV管径和密度较高。制模后14d、28d,A、B、C组炎性细胞密度较D组均显著减少(均为P<0.01);在制模后7d、14d、28d,A、B、C组较D组VEGF表达明显减少;A、B、C三组进行组间比较,仅制模后28d时B、C组间差异均无统计学意义(P>0.05),其余差异均有统计学意义(均为P<0.01);制模后7d、14d、28d,A、B、C三组均较D组CNV面积减少,差异均有统计学意义(均为P<0.01),而三组组间差异无统计学意义(P>0.05)。结论 不同浓度的KH902对兔碱烧伤后的CNV均有抑制作用,低浓度的KH902即可产生明显的抑制作用。

Abstract:Objective To investigate the inhibitive effects of different concentrations of KH902 eye drops on corneal neovascularization ( CNV) after rabbit corneal alkali burn. Methods Twenty-four adult rabbits were randomly divided into four groups , alkali bum model was established and divided into four groups, group A was treated with 2. 5 mg . mL-] KH902 eye drops, group B with 5 mg . mL-l KH902 eye drops , group C with 10 mg . mL -l KH902 eye drops , and control group with saline solution.3 times per day. At postoperative 3 days ,7 days ,14 days ,28 days , anterior segment photos were captured, the area of CNV was calculated, the confocal microscopy and pathological exanunation were given, and the vascular endothelial growth factor (VEGF) was analyzed quantitatively. Results CNV could be seen in four groups within 28 days , especially in group D from anterior segment photos. HE staining showed that the corneal epithelial and stromal damage and repair ,inflammatory cell infiltration and CNV grow were seen in four groups , but the inflammatory cells were more in group D , and the cube diameter and density of CNV were higher. At postoperative 14 days ,28 days ,the inflammatory cell density in group A, B , C were obviously lower than that in group D ( all P < 0. 01 ) . At postoperative 7 days , 14 days ,28 days , VEGF concentration in group A, B , C were obviously lower than that in group D ( all P < 0. 01 ) ; There were significant differences among group A, B, C ( all P < 0. 01 ) , except for 28 days between group B and C (P > 0. 05 ) . At postoperative 7 days, 14 days , 28 days , the CNV area in group A,B,C were obviously lower than that in group D ( all P < 0. 01) , and there was no statistical difference among group A, B, C ( P > 0. 05 ) . Conclusion KH902 eye drops with different concentrations have inhibitive effects on CNV after rabbit corneal alkali burn . even with lower concentration.
