摘    要:目的研究小鼠血小板/T细胞活化抗原1(PTA1)在小鼠组织及各种细胞系的表达分布与诱导,及其在杀伤性T细胞和LAK细胞诱导中的作用。方法应用流式细胞仪技术分析PTA1在小鼠组织及细胞系中的表达及诱导;用同种异体抗原诱导杀伤性T细胞,用IL-2诱导LAK细胞,分别研究PTA1特异性单克隆抗体对其分化的影响。结果小鼠PTA1主要在T细胞系呈诱导性表达,参与杀伤性T细胞的分化,但不参与LAK细胞的诱导。结论小鼠PTA1与人PTA1有相似的分布特点与功能,是一种进化中高度保守的白细胞分化抗原

Abstract:Objective To study the expression and function of murine platelets and T cell activation antigen 1 (PTA1), a novel leukocyte differentiation antigen initially described in human activated T cells and platelets , later in monkey and gibbon cells. Methods By using PTA1 specific McAb LeoA1, flow cytometry analysis of murine cell lines and murine mixed lymphocyte reaction (MLR) were taken. Results Flow cytometry analysis showed that murine PTA1 was expressed mainly in activated T cells, and this expression could be induced by phytohemoagglutinin (PHA) and phorbol ester (PMA). MLR experiment showed that LeoA1 McAb could block the induction of cytotoxic T lymphocytes (CTL) induced by alloantigen, but had no effect on the function of CTL when added at effector phase. LeoA1 had effect on neither the differentiation nor the function of lymphokine activated killer (LAK) cells. Conclusions PTA1 is a highly conserved leukocyte differentiation antigen during evolution, and murine PTA1 is a novel mouse leukocyte differentiation antigen with the same pattern of expression and function as human PTA1.
