连接蛋白37基因多态性与缺血性脑卒中和临床分型及预后

摘    要:目的探讨连接蛋白37(Cx37)基因C1019T多态性位点与缺血性脑卒中及其临床分型、预后的关系。方法选择缺血性脑卒中(脑卒中组)232例,又分为3个亚型组;动脉粥样硬化性血栓性脑梗死组(脑梗死组)115例、脑栓塞组31例、腔隙性脑梗死组(腔梗组)86例;另选健康者235例为对照组。采用限制性片段长度多态性分析技术,检测各组Cx37基因C1019T多态性分布。结果脑卒中组及脑梗死组TT基因型和T等位基因频率明显高于对照组(P<0.05)。多因素logistic回归分析显示,校正性别、年龄、体重指数、吸烟、空腹血糖、血压、血脂等因素后,Cx37基因C1019T的TT基因型脑卒中其亚型动脉粥样硬化性血栓性脑梗死发病风险明显增加(P<0.05)。但其各基因型与脑卒中预后无相关性(P>0.05)。结论 Cx37基因C1019T的T等位基因是缺血性脑卒中及其亚型动脉粥样硬化性血栓性脑梗死的遗传易感基因之一。Cx37基因多态性与缺血性脑卒中预后无关。

Polymorphism of connexin 37 gene and typing and prognosis of ischemic cerebral stroke

Abstract:Objective To study the C1019T polymorphisms of connexin 37(Cx37) gene and the typing and prognosis of ischemic cerebral stroke.Methods Two hundred and thirty-two patients with ischemic cerebral stroke served as a cerebral stroke group which was furhter divied into cerebral infarction group(n = 115 ),cerebral embolism group(n=31),lacunar infarction group(n = 86),and 235 healthy individuals served as a control group.C1019T polymorphisms of the Cx37 gene were detected by restriction fragment length polymorphism analyzing technique.Results The C1019T polymorphisms of the Cx37 gene were mainly manifested as CC genotype and C allele but rarely as TT genotype and T allele in different groups.The frequency of TT genotype and T allele was significantly higher in cerebral stroke group and cerebral infarction group than in control group(P<0.05).Multivariate logistic regression analysis showed that the risk of developing atherosclerotic and thrombotic cerebral infarction was significantly higher in patients with TT genotype of the Cx37 gene after adjustment of their gender,age,BMI,smoking,fasting blood sugar level and hypertension(P<0.05).However,no significant correlation was found between different genotypes of the Cx37 gene and prognosis of ischemic cerebral stroke(P>0.05).Conclusion The T allele of the Cx37 gene is one of the genetic genes susceptible to ischemic stroke and atherosclerotic and thrombotic cerebral infarction.C1019T polymorphisms of the Cx37 gene are not related with the prognosis of ischemic stroke patients.
