骨性关节炎大鼠软骨与滑膜基因表达的cDNA微阵列实验研究

摘    要:目的 研究骨性关节炎大鼠滑膜及软骨基因表达谱,探讨骨性关节炎与各种基因的相关性.方法 选取12只Wistar种大鼠,随机分为骨性关节炎模型组和正常对照组,每组6只,骨性关节炎模型组进行骨性关节炎造模.采用cDNA微阵列技术,根据遗传学中心法则,利用基因互补配对原理,用含有588个基因的大鼠基因表达谱芯片从基因表达水平在同一载体上同时进行多基因检测,研究骨性关节炎模型组及正常对照组大鼠滑膜基因表达谱,动态、整体、定量地考察骨性关节炎发生发展过程中基因种类、数量的改变. 结果 骨性关节炎模型组12周时可见关节软骨失去光泽,呈灰黄色,软骨变薄、碎裂,滑膜充血水肿.正常对照组未出现明显的骨性关节炎表现.与正常对照组大鼠软骨及滑膜的基因表达谱相比,骨性关节炎模型组大鼠软骨与滑膜中的差异表达基因共有82个,主要基因种类包括生长因子类基因、免疫基因、细胞凋亡基因、生长发育基因、疾病相关基因、能量相关基因及其他基因;其中上调基因27个,下调基因55个. 结论 本研究在骨性关节炎大鼠软骨与滑膜中检测出的82个差异表达基因可能与骨性关节炎的发生相关,为进一步基因治疗骨性关节炎提供了重要讯息.

Gene expression profile in synovium and cartilage of osteoarthritic rats: a cDNA microarray analysis
Abstract:Objective To explore the correlation between osteoarthritis and the gene expression profile in synovium and cartilage of osteoarthritie rats. Methods Twelve Wistar rats, divided into 2 even groups, were raised for 3 months to create an osteoarthritis group and a normal group, eDNA microarray chips containing 588 rat target genes were used to explore the gene expression profile in synovium and cartilage of both osteoarthritic and normal rats. The changes in the type and number of target genes at the onset and de-velopmental phases of osteoarthritis were observed dynamically, globally and quantitatively. Results Compared with the normal rats, of the genes expressed in the cartilage and synovium of osteoarthritie rats, 82 were identified to express differentially in osteoarthritie rats. Of them, 27 genes were up-regulated and 55 genes were down-regulated. Conclusion Osteoarthritis may be related to the abnormal gene expressions screened from the target, which can provide important information for further study of the osteoarthritis.
