新型分子生物学方法快速诊断眼库供体角膜病毒感染

目的应用多引物PCR方法快速诊断眼库供体角膜疱疹病毒感染，探讨角膜移植术后移植衰竭和疱疹病毒感染的关系。方法随机选择60例眼库供体角膜，54例拟接受角膜移植手术病人，术前应用多引物PCR趣方法同时检测HST-Ⅰ、HSV-Ⅱ、CMV、VZV。结果2例供体角膜（3．3％2／60）为HST-Ⅰ阳性，3例临床诊断为病毒性角膜炎病人2例查到HSV-Ⅰ（3．7％2／54），1例为VZV阳性（1．9％1／54）。结论多引物PCR方法是适合供体角膜特点的快速精确的诊断方法。

Multiplex polymerase chain reaction for detection of Herpes simplex virus type 1,type 2,cytomegalovirus and varicella-zoster virus in eye bank cornea donor

Objective To study the value of multiplex polymerase chain reaction(PCR) in diagnosing herpes virus infection of eye bank cornea donor.The correlation of primary graft failure(PGF) and herpes virus was discussed.Methods Sixty eye bank donors and 54 recipients were selected randomly before operation,and herpes simplex virus type 1(HSV-I),type 2(HSV-II),cytomegalovirus(CMV) and varicella-zoster virus(VZV) were detected by multiplex PCR.Results Two(3.3%,2/60) of the 60 donors in eye bank were HSVI positive.Three of 54 recipients who clinically diagnosed as viral keratitis were multiplex PCR positive,among them two recipients(3.7% 2/54) were HSV-I positive and one recipient(1.9% 1/54) was VZV positive.Conclusion Multiplex PCR is an accurate and a reliable method suits for eye bank donor to simultaneously detect several kinds of virus infection
