膜联蛋白A1表达下降在口腔鳞癌中的意义

摘    要:目的:研究膜联蛋白A1(ANXA1)在口腔鳞癌中的表达及其与临床病理的关系。方法:应用实时定量PCR、Western印迹方法和免疫组化方法检测ANXA1在口腔鳞癌细胞株和30例原发口腔鳞癌中的表达,采用SPSS10.0软件包对数据进行非参数检验。结果:与人永生化口腔黏膜上皮细胞(HIOEC)相比,Tca8113、TSCC、OSC和NT细胞中ANXA1的mRNA表达降低;Tca8113、TSCC、CAL-27、OSC和NT细胞中ANXA1的蛋白表达降低。30例口腔鳞癌组织标本中,ANXA1的mRNA和蛋白表达均较癌旁组织降低(p〈0.001)。ANXA1的表达水平与肿瘤的病理分化程度有关(mRNA:P=0.007;蛋白:P=0.006),与肿瘤大小、临床分期、淋巴结转移无关。结论:ANXA1在口腔鳞癌中表达降低,可能与肿瘤的发生、发展和组织分化有关。

Abstract:PURPOSE: To investigate the expression of Annexin A1 (ANXA1) in OSCC as well as its correlation with clinical pathology. METHODS: The mRNA level and protein expression of ANXA1 in some OSCC cell lines and human primary OSCC tissue specimens were detected using real-time PCR ,Western blotting and immunohistochemistry. The data was analyzed with SPSS10.0 software package for non-parametric analysis. RFSULTS: Decreased mRNA level and protein expression of ANXA1 were observed in some OSCC cell lines compared with the HIOEC. Decreased immunoreactions and mRNA levels of ANXA1 were also observed in OSCC tissue specimens compared with normal adjacent tissues. Moreover, ANXA1 expression was correlated with the tumor differentiation stage. CONCLUSIONS: The results suggest that decreased expression of ANXA1 contributes to cancerous progression and differentiation, and ANXA1 may be potentially used as a biomarker and a therapeutic target for OSCC.
