正电子发射计算机断层显像/18F-多巴胺受体显像和血浆18F-多巴放射性测定早期诊断帕金森病的实验研究

摘    要:目的:从受体水平上为早期诊断和研究帕金森病(Parkinsondisease,PD)提供依据。方法:对2只正常对照猴和12只MPTP猴进行正电子发射计算机断层显像(positronemisiontomography,PET)脑多巴胺受体显像,并在注药后不同时间测定血浆未代谢18F多巴。结果:MPTP猴不论有无锥体外系症状或体征,PET受体影像纹状体放射性分布较对照组皆明显减淡或缺损,示踪剂在纹状体特异性结合时间放射性曲线呈明显减低。在各个时间点测得血浆未代谢18F多巴皆高于对照组。结论:PET/18F多巴胺受体显像和血浆18F多巴放射性测定可用于临床型和亚临床型PD及其同类疾病的早期诊断和研究

Abstract:Objective: To provide evidence for the early diagnosis and research for Parkinson disease (PD) from the receptor level. Methods: 2 controls and 12 1 methyl 4 phenyl 1,2,3,6 tetrahydropyridine (MPTP) treated monkeys were performed with positron emission tomography (PET) cerebral dopamine receptor imaging, and the unchanged 18 F dopa in plasma was measured at different intervals after administration. Results: In MPTP treated monkeys with or without the development of extrapyramidal signs and symptoms, the data obtained with PET receptor imaging demonstrated an evident diminution or defect of 18 F dopa radioactive accumulation on striatum compared with the controls, and an apparent reduction of specifically bound radiotracer on striatum time radioactivity curve. The unchanged 18 F dopa in plasma determined at various intervals in MPTP treated monkeys was higher than that in the controls. Conclusion: The proposed approach can be used in the early diagnosis and research for clinical and subclinical PD and the similar disorders as well.
