摘    要:目的 建立HPLC法同时测定清肺抑火片中栀子苷、黄芩苷、大黄素和大黄酚的含量.方法 采用Waters C18色谱柱(4.6 mm ×250 mm,5 μm),以乙睛(A)-0.2%磷酸(B)为流动相梯度洗脱,流速1.0 mL·min-1,采用DAD检测器在200~400nm波长处进行检测,柱温30℃.结果 调取不同波长的色谱图分别计算各成分的含量,栀子苷(237 nm)、黄芩苷(277 nm)、大黄素(254 nm)和大黄酚(254 nm)进样量分别在0.2825~5.650μg(r2=1);0.719~14.383 μg(r2=0.9999);0.0415~0.83 μg(r2 =0.9998);0.0440~0.8795 μg(r2=0.9999)范围内呈现良好的线性关系.平均回收率(n=6)分别为97.93%,97.89%,97.91%,98.04%;RSD分别为0.14%,0.47%,0.45%,0.21%.结论 该方法快速简便,结果准确可靠,为清肺抑火片的全而质量控制提供科学的依据.

Simultaneous Determination of 4 Components in Qingfeiyihuo Talets by HPLC-DAD

Abstract:Objective: To establish a HPLC method for the determination of geniposide, baicalin, emodin and chrysophanol in Qing- feiyihuo Tablets. Methods: Waters C18column (4.6mm × 250mm, 5μm) was used with acetonitrile (A), -0.2% acid (B) as the mobile phase gradient elution, flow rate 1 mL · min-1 and the DAD detector was used to detect wavelength at 200 - 400 nm, with 30℃ of column temperature. Results: The different wavelength chromatograms were used to calculate geniposide (237nm), baicalin (277 nm) , emodin (254 nm) and chrysophanol (254 nm). The sample size showed a good linear relationship in the range of 0.2825 and 5. 650 μg ( r2 = 1 ), 0.719 - 14. 383μg ( r2 = 0. 9999), 0.0415 - 0.83 μg ( r2 = 0. 9998) and 0. 0440 - 0. 8795 μg ( r2 = 0. 9999). The average recovery rate (n =6) were 97.93%, 97.89%, 97.91%, 98.04% respectively, RSD = 0. 14%, 0.47%, 0.45%, 0.21% respectively. Conclusion: The method is simple, accurate and reliable and can provide the scientific basis for the whole quality control of Qingfeiyihuo Tablet.
