AG490影响电针足三里干预大鼠T细胞JAK/STAT信号转导途径时效关系研究

摘    要:目的:探讨氰基苄苯乙烯胺(AG490)影响电针足三里干预大鼠T细胞Janus酪氨酸激酶/信号转导和转录活化因子(JAK/STAT)信号转导途径的时效关系。方法:选用健康成年SpragueDawley雄性大鼠50只,随机分为对照组、足三里穴组、AG490短期组、AG490中期组、AG490长期组。应用流式细胞仪技术,通过免疫荧光染色法测定外周血T淋巴细胞亚群。结果:足三里穴组大鼠CD4+细胞百分率明显高于对照组(P〈0.01),CD8+细胞百分率与对照组相比无统计学意义(P〉0.05)。AG490长期组大鼠CD4+细胞百分率明显低于足三里穴组(P〈0.01),AG490中期组大鼠CD4+细胞百分率低于足三里穴组(P〈0.05),AG490短期组大鼠CD4+细胞百分率与足三里穴组相比无统计学意义(P〉0.05)。结论:电针足三里穴可明显提高正常大鼠的T淋巴细胞亚群,AG490可抑制电针足三里穴对T淋巴细胞亚群的调节作用,其抑制效应的强度在一定范围内与AG490的给药时间长度呈正相关。

Abstract:Objective:To observe the time-effect relationship of cyanobenzyl amine styrene(AG490) on JAK-STAT signalling pathway of T-lymphocyte in rats treated by electroacupuncture(EA) at Zusanli. Methods:Fifty Sprague Dawley rats were randomly divided into control group,Zusanli group,short period AG490 group,middle period AG490 group and long period AG490 group. T-lymphocyte subgroups of peripheral blood(including CD4+ and CD8+) were measured by direct microvolume whole blood immunoflurescent staining technique with flow cytometer(FCM) evaluate the immune function of rats from different aspects. Results:The value of CD4+ in Zusanli group was signifincantly higher than that of control group(P0.01),and the values of CD8+ between the two groups had no statistical difference(P 0.05). The value of CD4+ in long period AG490 group was significantly lower than Zusanli group(P 0.01),the value of CD4+ in middle period AG490 group was lower than Zusanli group(P0.05),the data of the short period AG490 group was not different from that of control group(P 0.05). Conclusion:EA on Zusanli point can improve cellular immune induced by T-lymphocytes,AG490 can inhibit the regulation effects on T-lymphocytes by EA on Zusanli point. Inhibitory effect was positively correlated with the times of using AG490 in a certain range.
