复方丹参注射液对血管内皮细胞基因表达谱的影响研究

摘    要: 目的探讨复方丹参注射液对血管内皮细胞基因表达谱的影响。方法培养人脐静脉内皮细胞;复方丹参注射液作用细胞18 h,以正常培养的细胞作对照,用心血管疾病及细胞因子相关的基因芯片研究复方丹参注射液对培养的人脐静脉内皮细胞基因表达谱的影响,并用RT-PCR方法对表达上调与表达下调的基因进行了扩增和电泳检测。结果基因芯片分析筛选出复方丹参对内皮细胞的影响基因9个,其中上调基因3个,下调基因6个,包括一些与免疫、血管舒缩、细胞黏附及抗氧化的相关基因;对差异表达的基因用RT-PCR扩增及电泳检测结果与基因芯片结果一致,证实了基因芯片的检测结果。结论复方丹参可通过多种途径在基因水平调节内皮细胞的功能。

Abstract:OBJECTIVE To investigate the effects of Fufang Danshen injection on the gene expression in the cultured human umbilical endothelial cells(HUVEC).METHODS HUVEC were cultivated and treated with Fufang Danshen injection(18 h).The alteration in the gene expression spectrum was analyzed using a gene microarray containing cardiovascular disease related genes and cytokine genes with the normally cultivated cells as control.The DNA micorarray in the differentially expressed genes was determined by RT-PCR.RESULTS 3 up regulated and 6 down regulated genes were identified after 18 h treatment of Fufang Danshen injection.These genes were involved in immunization,vasomotion,cell adhesion and anti-oxidation.And the result of RT-PCR was in accordance with that of the DNA microarray. CONCLUSION Fufang Danshen Injection regulates the human umbilical endothelial cells function at gene level through multiple pathways.
