氯胺酮对大鼠全脑缺血再灌注损伤的预防与治疗作用

摘    要:目的 观察氯胺酮对全脑缺血再灌注损伤的保护作用。方法 采用改良Pulsinelli 4血管法建立大鼠全脑缺血模型,缺血时间为15min。32只SD大鼠随机分假手术、缺血再灌注、氯胺酮治疗及氯胺酮预注4组。于动物存活第6天行Y-型迷宫检测大鼠的学习能力,第7天检测记忆能力并行海马CAl区组织病理学检查。结果 大鼠缺血再灌注后学习和记忆能力明显下降,海马CAl区大量神经元细胞坏死,氯胺酮治疗组与氯胺酮预注组能不同程度减轻海马CAl区神经元的损伤,改善缺血再灌注后学习和记忆的障碍。结论 氯胺酮对全脑缺血再灌注损伤有预防和治疗作用。

Abstract:Objective To investigate the effects of ketamine on neuronal injury following global ischemia and reperfusion in rats.Methods Global ischemia was induced by 15 minutes using the method described by Pulsinelli with modification.All rats were divided into 4 groups:sham-operated group;ischemia-reperfusion group;postischemia ketamine-treated group;preischemia ketamine-treated group.Y-type maze was used to detect the behavior change at day 6 and 7 postischemia.Histopathology test were carried out at day 7 postischemia.Results The learning and memory function of the rats were reduced after global ischemia and reperfusion.A large number of neurons lost in the CA1 region of hippocamus.More neurons survived in postischemia ketamine-treated group and preischemia ketamine-treated group.The learning and memory function were improved.Conclusion Ketamine provided neuroprotective effects.
