吲哚美辛对原代培养胃癌细胞端粒酶活性的调节作用

摘    要:目的:探讨吲哚美辛对原代培养胃癌细胞端粒酶活性的影响,研究其抗肿瘤的作用机制。方法:常规组织块培养法进行胃癌原代细胞培养,取终浓度为200、400及600μmol/L的吲哚美辛作用于原代培养胃癌细胞后,用MTT比色法测定细胞的生长抑制率,采用半定量-TRAP银染法检测端粒酶活性,流式细胞学检测Bcl-2表达水平。结果:不同浓度的吲哚美辛对原代培养胃癌细胞的增殖均有抑制作用,与对照组相比差异有统计学意义(P0.05);对胃癌细胞的端粒酶活性有抑制作用,其作用呈时间-剂量依赖性;端粒酶活性的降低与Bcl-2水平下降有关。结论:吲哚美辛能抑制胃癌细胞的增殖,降低端粒酶活性,作用机制与Bcl-2水平下降有关。

Abstract:Objeetive: To study the effects of indomethacin, a COX-2 inhibitor ,on cell prolif- eration, telomerase activity in primary cultured gastric cancer cells and to explore its anti-neoplasm mechanism. Methods: M-FF assay was used to determine cell proliferation after incubation in dif- ferent concentrations (200,400,600 μmol/L) of indomethacin. Telomerase activity was detected by semi-TRAP assay. Flow cytometry was adopted to examine Bcl-2 level. Results: Indomethacin inhibited significantly the growth of primary cultured gastric cancer cells and elevated the inhibiting rate (P〈0.051)as compared with control groups. The telomerase activity was significantly inhibited in dose and time dependent manner, and the inhibition of telomerase activity was associated with de- crease of Bcl-2 level. Conclusions: COX-2 inhibitor indomethacin inhibited proliferation and inhib- ited telomerase activity by partly decreasing Bcl-2 level in primary cultured gastric cancer cells. The results indicate one of the mechanisms underlying the anti-cancer effect of COX-2 inhibitor.
