活血通脉汤对大鼠脑缺血再灌注损伤诱导的凋亡蛋白Fas、Caspase-3表达的影响

摘    要:目的探讨活血通脉汤对大鼠脑缺血再灌注损伤过程中所诱导的凋亡蛋白Fas、Caspase-3表达的影响。方法制作脑缺血再灌注模型,80只大鼠随机平均分为4组,假手术组、模型组、阿司匹林组、活血通脉汤组,每组20只。应用免疫组织化学方法分别检测每组大鼠脑缺血再灌注12、24h脑组织Fas、Caspase-3表达的情况。结果模型组大鼠脑缺血再灌注12、24h脑组织Fas、Caspase-3表达水平明显高于假手术组(P〈0.01);活血通脉汤组大鼠各时间点脑组织Fas、Caspase-3表达水平明显低于模型组(P〈0.05,P〈0.01),且与阿司匹林组无显著差异性(P〉0.05)。结论活血通脉汤能降低大鼠脑组织Fas、Caspase-3表达水平,减轻缺血脑组织神经元坏死的程度,对大鼠脑缺血再灌注损伤具有保护作用,机制与降低Fas、Caspase-3表达水平有关。

Abstract:Objective To investigate the impact of Huoxuetongmai decoction on the expression of Fas, Caspase-3 induced by cerebral ischemia reperfusion injury in rats. Methods 80 rats were randomly divided into four groups: sham operated group( n = 20), model group( n = 20), aspirin group( n = 20), Huoxuetongmai decoction group ( n = 20). 24h after ischemia reperfusion, rats were executed . The area of cerebral infarction and the pathological change in ischemia area were observed by HE staining. The expression levels of Fas, Caspase-3 were measured by immunohistochemistry. Results In comparison with sham operated group, the area of cerebral infarction obviously increased, the expression of Fas and Caspase-3 in brain tissue obvious- ly increased at 12h and 24h after reperfusion in model group (P 〈0.01) . In comparison with model group, the area of cerebral infarction obviously reduced and the expression of Fas, Caspase-3 significantly decreased in Huoxuetongmai decoction group (P 〈 0.01, P 〈 0.05). There was no difference between Huoxuetongmai decoction group and aspirin group( P 〉 0. 05). Conclusion Huoxuetongmai decoction could inhibit excessive expression of Fas and Caspase-3, thereby relieve cerebral ischemia reperfusion injury.
