摘    要:目的:探讨白介素10(IL-10)对伴放线放线杆菌内毒素(Aa—LPS)体外诱导兔肺巨噬细胞凋亡作用的影响。方法:经兔气管肺泡灌洗获得肺泡巨噬细胞,随机分为空白对照组、Aa—LPS组、Aa—LPS+IL-10组。按实验分组加入Aa—LPS(1汕g/mL)、IL-lo(o.1p.g/mL),24h后裂解细胞,荧光定量PCR法检测促凋亡基因bax、p53和caspase-3的表达。结果:Aa—LPS组bax、caspase-3的表达较空白对照组明显升高(P〈0.05),p53的表达与空白对照组比较差异无统计学意义(P〉0.05)。Aa—LPS+IL-10组bax、p53、caspase.3的表达较Aa—LPS组降低(P〈0.05)。结论:Aa—LPS体外对肺巨噬细胞有促凋亡作用,IL-10可抑制Aa—LPS的促凋亡作用,其机制可能与细胞凋亡的线粒体途径有关。

Abstract:Objective: To investigate the effect of Aa-LPS and IL-10 on apoptosis of rabbit alveolar macrophages (AM) in vitro. Methods: AM were harvested from the bronchoalveolar lavage fluid. These cells were randomly divided into three groups: control group,Aa-LPS group and Aa-LPS + 1L-10 group. Then these cells were given Aa-LPS (1 μg/mL) ,and IL-10 (0.1 μg/mL). After 24 hours,the cells were harvested to extract RNA. Finally, real-time RT-PCR was used to detect the expression of gene bax, p53 and caspase-3. Results :The results showed that the expression of bax and caspase-3 in the Aa-LPS group was significantly higher than that of the control group (P 〈 0.05 ). The expression of p53 was not statistically significant compared with that of the control group (P 〉 0.05 ). IL-10 treatment significantly inhibited macrophagocyte apoptosis and suppressed the expression of bax ,p53 and caspase-3 ( P 〈 0.05). Conclusions:Aa-LPS can promote alveolar macrophages apoptosis in vitro and IL-10 can suppress the effect of Aa-LPS. The mechanism seemed related to the mitochondria-dependent pathway.
