高效液相色谱法测定瓜蒌薤白滴丸中乙酸乙酯部位槲皮素的含量及特征图谱

目的:建立测定瓜蒌薤白滴丸乙酸乙酯部位槲皮素含量的高效液相色谱(HPLC)分析方法,并在此基础上建立乙酸乙酯部位的特征图谱。方法:通过HPLC-PDAD联用建立瓜蒌薤白滴丸乙酸乙酯部位特征图谱,测定槲皮素的含量;利用均值和中位数建立共有模式,采用相关系数和夹角余弦计算瓜蒌薤白滴丸乙酸乙酯部位与共有模式间的相似程度。结果:槲皮素在0.10².00μg范围内线性关系良好,回归方程为:y=9 812.2x+2 433.5,相关系数r=0.999 8,平均回收率为98.8%,RSD(%)=5.52%,13个批次瓜蒌薤白滴丸乙酸乙酯部位槲皮素的平均含量为0.014%,RSD(%)=10.2%。建立了瓜蒌薤白滴丸乙酸乙酯部位HPLC特征图谱,取共有峰的均值和中位数,利用相关系数法计算平均相似度,分别为0.888 4和0.873 9;利用夹角余弦法计算相似度,分别为0.921 7和0.913 1。结论:瓜蒌薤白滴丸乙酸乙酯部位中槲皮素的含量测定方法的重复性及稳定性较好,同时建立了瓜蒌薤白滴丸乙酸乙酯部位特征图谱,可用于瓜蒌薤白滴丸乙酸乙酯部位的质量评价。

Determining quercetin content and characteristic spectrum of ethyl acetate extraction in Gualou Xiebai pills by HPLC

Objective：To establish an analysis method for determining quercetin content of ethyl acetate extraction in Gualou Xiebai pills by high-perform-ance liquid chromatography（HPLC）,on which basis to develop the characteristic spectrum for ethyl acetate extraction.Methods：The characteristic spec-trum for ethyl acetate extraction in Gualou Xiebai pills was developed with HPLC-PDAD,and the quercetin content was measured .The common pattern was established on the mean and the median basis,and the similarity degree between ethyl acetate extraction in Gualou Xiebai pills and the common pattern was calculated by the correlation coefficient method and the included angle cosine method .Results：A good linear relationship for quercetin content was repre-sented at range of 0.10-2.00 μg by the regression equation of y =9 812.2x ＋2 433.5（r=0.999 8）,which had led to the average recovery rate of 98.8% and relative standard deviation（RSD） as 5.52%.The average content of quercetin and its RDS was 0.014% and 10.2%,respectively,deter-mined in 13 different batches of ethyl acetate extraction in Gualou Xiebai pills.The characteristic spectrum of ethyl acetate extract in Gualou Xiebai pills was successfully established.The average similarity degree was 0.888 4 and 0.873 9,0.921 7 and 0.913 1,respectively,calculated by both correlation co-efficient and included angle cosine methods on the mean and median of common peaks .Conclusion：This method can be reproductive and stable in deter-mining the quercetin content of ethyl acetate extraction in Gualou Xiebai pills.Successfully established common pattern for the characteristic spectrum of ethyl acetate extraction in this agent together with the previous procedures may contribute to better quality evaluation for the ethyl acetate extraction in Gua-lou Xiebai pills.
