小鼠大肠上皮细胞恶性转化中细胞唾液酸粘蛋白的分布及意义

摘    要:采用N-甲基-N-亚硝基脲(MNU)直肠灌注诱发小鼠实验性大肠癌,以高铁双胺-奥蓝(HID-AB)粘蛋白组织化学方法观察诱癌过程中大肠上皮细胞唾液酸粘蛋白组织化学改变及其与大肠上皮细胞形态恶性转化的关系,结果表明第6周起小鼠远端远端结肠呈唾液酸粘蛋白反应上皮细胞数量显著增加,蓝染的唾液酸粘酸粘物质多位于细胞浆,增生的腺腔缘及腺腔分泌物,起始于大肠上皮细胞形态转化之前,峰值出现于原位癌及浸润癌形成前

Abstract:The sequential histological and acidic mucin hlstochemical changes in colonic epithelium during carcino-genesis were studied in female ICR mice, which were given l2 weekly intrarectal instilla- tion of N-methyl-N-nitrosourea(MNU ). The mucin histochemical changes were assessed by the use of high iron diamine-alcian blue(pH 2. 5)staining. In the untreated controls, the epithelial cells in the distal colon were characterized by a predominance of sulfomucins, particularly in the upper crypt and surface cells, with occasional sulfomucins in the base of the crypt. The earlier mucin histochemical changes de- tected at the 6th week were characterized by the progressively increased staining of sialomucins and ac- companied in general by a decrease or absence of sulfomucins, similar to the changes described in the tran- sitional mucosa(TM ) adjacent to colorectal cancer in the human. Sialomucin staining occurred mainly in the cytoplasm and the luminal surface of the crypt. The results suggested that the malignant transforma- tion of colonic epithelium was related to qualitatively enhanced sialomucins in concert with reduced sulfo- mucin staining, Detection of sialomucins might be a simple and reliable screening technique in the early diagnosis of premalignant change of colon.
