绞股蓝总甙对大鼠心、脑微粒体Na~+,K~+-ATP酶活性的抑制作用

摘    要:本文对照人参总甙观察了绞股蓝总甙对大鼠心、脑微粒体Na ̄+,K ̄+-ATP酶活性的影响。结果表明:绞股蓝总甙体外可逆性抑制鼠心、脑微粒体ATP酶,并具有浓度依赖性,IC_(50)分别为58.79±8.05mg/L和52.07±6.25mg/L。Na ̄+,K ̄+对绞股蓝总甙的抑酶作用有拮抗。酶动力学分析显示:绞股蓝总甙对微粒体Na ̄+,K ̄+-ATP酶的底物ATP的抑制为反竞争性抑制。结果提示:绞股蓝总甙的强心和中枢抑制作用可能与对心、脑Na ̄+,K ̄+-ATP酶的抑制作用有关。

Abstract:Comparing with ginsenoside (Gin ). the effects of gypenoside(Gyp)on themicrosomal Na ̄+,K ̄+-ATPase from rat hearts and brains were investigated.The resultsshowed that Gyp inhibited the enzyme rapidly and reversibly in vitro. The inhibition was in aconcentration- dependent manner. The IC_(50) of Gyp for the heart and brain was 58.79 + 8.05and 52.07±6.25 mg/L,respectively.The inhibition was enhanced with increasing the con-centration of ATP or decreasing that of Na ̄+ and K ̄+. Kinetic analysis revealed that Gyp wasan uncompetitive inhibitor with respect to ATP. These results suggested that the positive in-otropic effect and the inhibition of CNS by Gyp are related to its inhibition on the activity ofNa ̄+,K ̄+-ATPase from hearts or brains.
