氯化钾除极对分离的豚鼠心室肌细胞胞浆内[Ca^2+]i的影响

摘    要:用酶解法分离单个豚鼠心室肌细胞,以荧光探针Fluo-3/AM标记胸浆内游离钙离子,用激光扫人聚焦显微镜实时测定胞浆内「Ca^+」变化。分别观察了在正常台氏液和无钙台氏液中30mmol/LKCl除极对胞浆内「Ca^2+」i的影响。

Abstract:To investigate the relationship between the membrane depolarization and the release of intracellular Ca 2 stores, single cardiac ventricular myocytes of guinea pig were isolated by an enzymatic technique and loaded with the Ca 2 indicator Fluo 3/AM. The changes of cytosolic Ca 2 ] i of ventricular myocytes depolarized by 30mmol/L KCl in normal Tyrode solution and Ca 2 free Tyrode solution were observed by using laser scanning confocal microscope. Results showed that depolarization by 30mmol/L KCl caused constant increase of cytosolic Ca 2 ] i during 250s in normal Tyrode solution and transient increase in Ca 2 free Tyrode solution. We concluded that depolarization caused the Ca 2 release from intracellular Ca 2 stores and Ca 2 influx was not required to mediate, the constant increase of cytosolic Ca 2 ] i was sustained by Ca 2 influx.
