摘    要:目的:优选聚酰胺树脂分离纯化绞股蓝总黄酮的工艺条件.方法:采用紫外分光光度法,以总黄酮含量为考察指标,研究在静态和动态条件下,聚酰胺树脂吸附和洗脱总黄酮的行为,筛选出聚酰胺树脂精制绞股蓝总黄酮的最优工艺参数.结果:最佳工艺参数为生药质量浓度0.35 g·mL-1,上样液pH 3,温度为室温,4 BV 60%乙醇洗脱.此工艺条件下,总黄酮纯度由粗品的23.91%提高到81.97%,纯化物中总黄酮保留率高达90.11%.结论:采用聚酰胺树脂纯化绞股蓝总黄酮效果良好,适用于工业化生产.

Abstract:Objective: To optimize purification technology for total flavonoids from Gynostemma pentaphyllum by polyamide resin. Method: With the content of total flavonoids as index,UV spectrophotometry was used, adsorption and elution behavior of polyamide resin for total flavonoids was investigated under static and dynamic conditions, and to select optimum technology parameters of purification technology for total flavonoids by polyamide resin. Result: Optimum purification technology parameters were:at room temperature,the concentration of crude drug 0.35 g·mL-1,pH 3.0 and 4 BV 60% ethanol as eluant. Under these conditions,purity of total flavonoids increased from 23.91% to 81.97%,retention rate of total flavonoids in the purified was up to 90.11%. Conclusion: Purified effect of total flavonoids from G. pentaphyllum by polyamide resin was preferable,it was suitable for industrial production.
