BCG激活膀胱肿瘤浸润淋巴细胞的抗肿瘤作用研究

为了探讨ＢＣＧ的抗肿瘤机理，从１５例手术切除的膀胱移行细胞癌新鲜组织标本中制备肿瘤浸润淋巴细胞（ＴＩＬ），分别在含ＢＣＧ或ＩＬ－２的全培养基中培养扩增，测定不同培养时间的抗瘤活性。结果：用活ＢＣＧ培养的ＴＩＬ第１２天对自体肿瘤细胞的杀伤活性达高峰，杀伤率为４８．３％；用ＩＬ－２培养的ＴＩＬ第１４天达杀伤高峰，杀伤率为４３．８％，用死ＢＣＧ培养的ＴＩＬ，其扩增结果和抗瘤活性均明显低于活ＢＣＧ及ＩＬ－２。提示ＢＣＧ对ＴＩＬ的直接激活作用可能是其抗肿瘤机理之一。

Antitumor activity of BCG activated tumor infiltrating lymphocytes on bladder tumor cells

Tumor infiltrating lymphocytes (TIL)were obtained from surgically resected fresh specimens from 15 cases of bladder transitional cell carcinoma, Stimulation of TIL was carried out by incubating in medium with viable BCG or with interleukin- 2(IL-2)and their antitumor activity was detemined by LDH release assay after different periods of incubation,The highest cytotoxicity of TIL incubated with viable BCG was about 48.3?ter 12 days of incubation while that of TIL incubated with IL-2 was about 43.8% after 14 days of incubation. The cytotoxicity effect of TIL incubated with dead BCG has been much inferior. It was suggested that the activated effect of viable BCG on suppressed TIL might be an important aspect of its antitumor mechanism.
