益气强心中药复方对不同动物离体心房肌收缩力和心搏频率的影响

目的观察益气强心中药复方对离体心房的心搏频率、心房肌收缩力的影响。方法取成年SD大鼠、豚鼠各40只,断头处死后,迅速打开胸腔摘取心脏,分离并截取相连的左右心房,标本悬挂于经95%O2和5%CO2饱和的Krebs-Henseleit液的浴槽中,其下端固定,上端连接张力换能器并施加一定的静息张力(0.5g),用超级恒温槽水浴循环维持在35.5℃左右,待心房可恢复自主跳动并平稳后开始进行实验,利用MS4000U-1C生物信号定量记录分析系统采集不同浓度的益气强心中药复方对心房肌收缩力和心搏频率的影响。结果益气强心中药复方能明显增加大鼠、豚鼠离体心房肌收缩力,25mg/L浓度时可使心肌收缩力增强1倍,在5～25mg/L浓度范围内呈剂量依赖性;益气强心中药复方能明显减慢大鼠、豚鼠离体心房心肌的收缩频率,在5～25mg/L浓度范围内呈剂量依赖性,并存在一定的动物种属差异。结论益气强心中药复方对动物离体心房有正性肌力和负性频率作用。

Effect of compound Chinese Traditional Medicine Yiqiqiangxin decoction on contractility and heart rate of isolated atrium in different animals

Objective To observe the effects of Yiqiqiangxin decoction on contractility and heart rate of isolated atrium in different animals.Methods The isolated atria from 40 SD rats,guinea pigs were prepared.Before and after giving Yiqiqiangxin decoction,the contractile force and heart rate were measured by MS4000U-1C biological signal collecting system.Results Yiqiqiangxin decoction could markedly enhance contractile force and inhibit heart rate of isolated atrium from rats and guinea pigs in a concentration-dependent manner.Conclusion Yiqiqiangxin Decoction has positive inotropic and negative chronotropic effects on isolated atrium from different species.
