巢蛋白在成年C57小鼠和SD大鼠中枢神经系统中的分布及比较

摘    要:
目的 观察巢蛋白(nestin)在成年C57小鼠和SD大鼠中枢神经系统中的分布,探讨并比较神经干细胞在两种啮齿类动物中分布的异同。方法 采用免疫组织化学ABC法和间接免疫荧光法,显示含神经干细胞特征性的标志物巢蛋白的阳性结构在上述两种啮齿类动物中枢神经系统中的分布。结果 在成年C57小鼠中枢神经系统中,巢蛋白在整个脑室系统周围均有表达,免疫阳性结构位于室管膜细胞胞体及其突起,并呈现由吻端向尾端渐次减少的趋势。在成年SD大鼠的脑室系统中,巢蛋白主要分布在侧脑室外侧壁及其周围脑区。此外,在两者的齿状回、穹窿下器、最后区等脑结构中也有巢蛋白的表达。成年C57小鼠中枢神经系统中巢蛋白的表达,无论在免疫阳性结构的数量上还是在染色强度上都明显多于和强于成年SD大鼠。结论 巢蛋白在成年C57小鼠中枢神经系统中的广泛脑区均有表达,而在成年SD大鼠中仅见于有限的脑区,提示成年C57小鼠中枢神经系统可能具有较强的神经可塑性和损伤修复能力。

Abstract:
Objective To observe the distribution of nestin in the central nervous system(CNS) of the adult Sprague-Dawley rats and C57 mice and to explore or compare the different location of neural stem cells between the two kinds of rodents. Methods Immunohistochemical and immunofluorecent methods were used to reveal the distribution of the immunoreactive structures containing nestin protein,a characteristic markers for neural stem cell,in the CNS of these two rodent species. Results In ths CNS of adult C57 mice,nestin immunoreactive products were mainly confined in the ependymal soma and their arborizations,which were seen in the whole cerebroventricular system and adjacent regions with a gradual decreased tendency from rostral to caudal.In the CNS of adult SD rats,however,nestin protein was mainly located in the lateral wall of the lateral ventricle and the adjacent region.In addition,the expression of nestin was also seen in the cells in dentate gyrus of the hippocampal structure,subfornical organ and the area postrema in both mice and rats.In general,the expression of nestin in the CNS of the adult C57 mice is much stronger than that in the adult SD rats.Conclusions Nestin is more widely expressed in the CNS in adult C57 mouse,but is only expressed in limited regions of the adult SD rat.The results of the present study indicate that C57 mice may have a more powerful ability for neural plasticity and the self-repair function after injury than adult SD rats.
